The first one that comes to mind is
Thelma and Louise (1991). Susan Sarandon and Gina David play a waitress and a housewife who take off across the country in their Thunderbird. Then Louise kills a man who has tried to rape Thelma, and now they're really on the run!

Tommy Lee Jones and Will Smith make good buddies in
Men in Black (1997). They try to save the world from aliens, then again, in the sequel
Men in Black II (2002).

Then there's one of the greatest comedies of all times,
Ferris Bueller's Day Off (1986). Alan Ruck's Cameron makes an excellent, if somewhat neurotic sidekick to Matthew Broderick's Ferris. High school senior Ferris decides to skip school (again) with the help of his sidekick and girlfriend. Funny movie!
